During aging, FABP7+ astrocytes develop cytoplasmic granules derived from degenerating mitochondria. Blood glucose levels can also be normalized in diabetic rodents by a single intrahypothalamic infusion of Fibroblast Growth Factor 1 (FGF1), an effect that persists for months even in severely diabetic animals. Hyperglycaemia, which is first detected during pregnancy, is classified as gestational diabetes mellitus (GDM), also known as hyperglycemia in pregnancy. Although it can occur anytime during pregnancy, GDM generally affects pregnant women during the second and third trimesters. This evidences the need to validate specific techniques using more accurate and sensitive techniques for determining glucose in unconventional fluids, like saliva [31, 32]. In 2015, Liu et al. described a dual-enzyme biosensor composed of glucose oxidase (GOx) and pistol-like DNAzyme (PLDz) with the image source ability to quantify glucose in tears and saliva [33]. Lee et al. in 2017 presented a wearable monitoring device for glucose in sweat, including a transdermal drug delivery module [34]. The study concluded that the device would require improvements for long-term stability and uniformity of sensors.
More in-depth studies of the relationship between blood glucose levels and sweat in healthy and individuals with diabetes are also needed. The same year, Soni et al. reported a portable, non-invasive optical glucose biosensor using a smartphone application to measure glucose in saliva and its clinical validation [35]. In 2019, de Castro et al. gave proof of concept for a wearable glucose monitor based on a microfluidic paper platform integrated into a mouth guard for point-of-care testing (POCT) [36].
Similar to the case in insulin-sensitive tissues, in states of IR, physiologic levels of circulating insulin are insufficient to elicit the appropriate insulin response in hepatic cells [176]. In the liver, IR impairs glycogen synthesis, fails to suppress glucose production, enhances lipogenesis, and increases the synthesis of proteins such as the proinflammatory CRP. In fact, the abnormal production of proinflammatory proteins such as adipocytokines and cytokines, combined with conditions such as oxidative stress, can lead to an inflammatory state responsible for altered insulin response by the liver [175]. Parallel mediation analysis was performed in participants with a duration of more than 7 years, including RC as the independent variable, DR as the dependent variable, and CRAE, CRVE, and Df as mediators (Fig. 2, Table 4). CRAE had no significant mediating effect on the relationship between RC and DR (0.003; 95% CI [-0.003, 0.011]). CRVE was a significant mediator of this association (0.020; 95% CI [0.012, 0.032]). Conversely, Df was a suppressor of this association (- 0.008; 95% CI [- 0.017, – 0.003]), but the suppressive effect was less pronounced than the mediating effect of CRVE. Therefore, the association between RC and DR is likely mediated by the diameter of the retinal venules.

P2Y purinoreceptors have been reported to be coupled to G-proteins [45,46] whereas P2X-type receptors are ATP-activated ligand-gated ion channels non-selective for cations [47].
